Performance Data
During experimental testing of the part, the part performed in the opposite manner than what was expected. As pH increased (increasing amount of lactic acid added to M9 that cells were grown in), the measured fluorescence produced by the cell decreased. Three experiments were run testing different dynamic ranges of acid added and all showed the same trend.
Another assay was run to ensure the part was not inducible by lactose. The data showed the part being induced greater at intermediate ranges and equally at low and high ranges (of a log scale). This would imply a more complex underlying transcriptional regulation system, an unlikely result given that our promoter is not endogenous to E. coli, and therefore unlikely to be incorporated into preexisting multi-component regulation systems. However the data does not prove that this part is not inducible by lactose so the research in this area is inconclusive.
More research needs to be done into either field in order to find the usefulness (or lack there of) of this part. Particularly, research could be made into some possible sources of experimental error such as growing the cells in a dark place so as to prevent acid "aging" (loss of potency over time due to the exposure to light), ensuring that growing the cells aerobically does not remove all lactic acid from the environment (in aerobic conditions, E. coli can utilize organic acids as a carbon source), or if E. coli is missing some factor that is present in L. lactis that will allow the part to function properly.
